Biyodizel, biyolojik olarak parçalanabilen, yenilenebilir ve toksik olmayan alternatif bir yakıttır. Biyodizel üretiminde temel hammaddenin bitkisel yağlar olması ve bu yağların özellikle tarım ürünlerinden elde edilmesi, biyodizel üretimini kısıtlamaktadır. Biyodizel üretiminde tarım ürünleri dışında farklı kaynakların kullanılması, hem biyodizel üretiminde devamlılığı sağlayacak hem de gıda bitkilerinin enerji sektöründe heba edilmesini önleyecektir. Bu çalışmanın amacı, çeşitli gıda örneklerinden izole edilmiş maya türlerinin lipit içeriği, lipit verimi ve yağ asidi profili bakımından biyodizel üretim potansiyellerini belirlemektir. Bu amaç doğrultusunda; çalışmada çeşitli gıda ürünlerinden izole edilen 8 adet maya izolatı, pH 5,5, 30 oC sıcaklık 100 rpm (çalkalama hızı) ve 24 saat sürede hem optimal besiyerinde (azot varlığında) hem de lipit üretim besiyerinde (azot yokluğu) inkübe edilerek lipit verimi ve % lipit miktarı bakımından karşılaştırılmıştır. Bu çalışmada en yüksek % lipit miktarı ve lipit verimine sahip olan B37 ve B53 kodlu izolatlar sırasıyla Pichia kudriavzevii, Saccharomyces cerevisiae türleri olarak DNA düzeyinde tanımlanmıştır. Tür tanımlaması yapılan Pichia kudriavzevii B37 ve Saccharomyces cerevisiae B53 suşlarına 30 oC sıcaklık 100 rpm ve 72 saat boyunca farklı pH (6-8-10) değerlerinde azot yokluğu stresi uygulanmıştır. Pichia kudriavzevii B37 kodlu izolatın pH 6'da % 34,4 olarak en yüksek % lipit miktarına, pH 8'de ise 261,37 mg/L/gün olarak en yüksek lipit verimine sahip olduğu görülmüştür. Saccharomyces cerevisiae B53 kodlu izolatın pH 10'da % 44,3 olarak en yüksek % lipit miktarına ve 730,60 mg/L/gün olarak en yüksek lipit verimine sahip olduğu görülmüştür. Maya hücrelerinden ekstre edilen lipitler transesterifikasyona uğratılarak elde edilen metil esterleri gaz kromotografisi-kütle spektrometresi kullanılarak analiz edilmiştir. Pichia kudriavzevii olduğu belirlenen B37 ve Saccharomyces cerevisiae olduğu belirlenen B53 kodlu izolatların kuru biyokütlesinden ekstre edilen lipitlerin transesterifikasyonu sonucunda sırasıyla % 96,02 ve % 96,28 oranında biyodizel verimi elde edilmiştir. Pichia kudriavzevii B37 ve Saccharomyces cerevisiae B53 kodlu izolatların yağ asidi dağılımı Pichia kudriavzevii B37 kodlu izolat için palmitik asit (% 15,61), palmitoleik asit (% 9,73), stearik asit (% 15,52), oleik asit (% 37,14) ve linoleik asit (% 14,96) olarak bulunmuştur. Saccharomyces cerevisiae B53 kodlu izolatta ise palmitik asit (% 12,00), palmitoleik asit (% 41,49), stearik asit (% 9,62) ve oleik asit (% 33,17) olarak bulunmuştur. Çalışmada kullanılan Pichia kudriavzevii ve Saccharomyces cerevisiae türlerinin yüksek lipit içeriğine ve lipit verimine sahip olmasının yanı sıra elde edilen lipitlerin büyük bölümünün C16 ve C18 metil esterlerinden oluşması bu mayaların sentezlediği lipitlerin biyodizel üretiminde hammadde olarak kullanılabilme potansiyeline sahip olduğunu göstermiştir.
Biodiesel is a biodegradable, renewable and non-toxic alternative fuel. The fact that the basic raw material in biodiesel production is vegetable oils and that these oils are obtained from agricultural products, limits the production of biodiesel. The use of different sources other than agricultural products in biodiesel production will both ensure continuity in biodiesel production and prevent food crops from being wasted in the energy sector. The aim of this study is to determine the biodiesel production potential of yeast strains isolated from various food samples in terms of lipid content, lipid yield and fatty acid profile. In accordance with this purpose; In this study, 8 yeast isolates isolated from various food products were incubated at pH 5,5, 30 oC temperature 100 rpm (shaking speed) and 24 hours both in optimal medium (in the presence of nitrogen) and in lipid production medium (lack of nitrogen) to increase lipid yield and Compared in terms of % lipid content. In this study, B37 and B53 coded isolates with the highest lipid content and lipid yield were identified at the DNA level as Pichia kudriavzevii and Saccharomyces cerevisiae species, respectively. Nitrogen deficiency stress was applied to Pichia kudriavzevii B37 and Saccharomyces cerevisiae B53 strains, which were identified as species, at 30 oC temperature, 100 rpm and different pH (6-8-10) values for 72 hours. Pichia kudriavzevii B37 coded isolate was found to have the highest lipid content of 34,4% at pH 6, and the highest lipid yield at pH 8 as 261,37 mg/L/day. It was observed that Saccharomyces cerevisiae B53 coded isolate had the highest lipid content of 44,3% at pH 10 and the highest lipid yield of 730,60 mg/L/day. The methyl esters obtained by transesterification of lipids extracted from yeast cells were analyzed using gas chromatography-mass spectrometry. As a result of the transesterification of lipids extracted from the dry biomass of B37 coded isolates, which were determined to be Pichia kudriavzevii and B53 coded isolates, which were determined to be Saccharomyces cerevisiae, biodiesel yields of 96,02% and 96,28% were obtained, respectively. Fatty acid distribution of isolates coded Pichia kudriavzevii B37 and Saccharomyces cerevisiae B53 For isolate coded Pichia kudriavzevii B37 palmitic acid (15,61%), palmitoleic acid (9,73%), stearic acid (15,52%), oleic acid (37%) ,14) and linoleic acid (14,96%). Palmitic acid (12,00%), palmitoleic acid (41,49%), stearic acid (9,62%) and oleic acid (33,17%) were found in Saccharomyces cerevisiae isolate coded B53. The fact that Pichia kudriavzevii and Saccharomyces cerevisiae species used in the study have high lipid content and lipid yield, as well as the fact that most of the lipids obtained are composed of C16 and C18 methyl esters showed that the lipids synthesized by these yeasts have the potential to be used as raw materials in the production of biodiesel.
