Mayalar bazı koşullar altında hücre içinde yüksek miktarda lipit üretimi yaptıkları için biyodizel üretiminde hammadde olarak kullanılabilme potansiyeli olan mikroorganizmalardır. Bu çalışmanın amacı çok az çalışılmış olan gıda kaynaklı mikroorganizmalardan mayaların biyodizel üretim potansiyellerini belirlemektir. Bu amaç doğrultusunda; izole edilen mayalar arasından azot yokluğu stresinde en yüksek lipit verimine ve biyodizel üretimi için uygun yağ asidi kompozisyonuna sahip maya izolatlarını keşfetmek hedeflenmiştir. Gıdalardan elde edilen 8 adet maya izolatı pH 5.5, 30 oC sıcaklık, 100 rpm (çalkalama hızı) ve 24 saat sürede hem optimal besiyerinde (azot varlığında) hem de lipit üretim besiyerinde (azot yokluğu) inkübe edilerek lipit verimi ve % lipit miktarı bakımından karşılaştırılmış olup en yüksek değerlere sahip olan A10 ve A20 kodlu izolatlar Pichia kudriavzevii türü olarak tanımlanmışlardır. Tür tanımlaması yapılan A10 ve A20 kodlu izolatlara 30 oC sıcaklık 100 rpm ve 72 saat boyunca farklı pH (6-8-10) değerlerinde azot yokluğu stresi uygulanmıştır. Her iki izolatın da pH 10'da en yüksek % lipit miktarına ulaştığı (A10 %40, A20 %27) görülmüştür. A10 kodlu izolat pH 10'da 431,86 mg/L/gün ile en yüksek lipit verimine ulaşırken, A20 kodlu izolat ise pH 6'da 219,37 mg/L/gün ile en yüksek lipit verimine ulaşmıştır. Maya hücrelerinden ekstre edilen lipitler transesterifikasyona uğratılarak elde edilen metil esterleri GC-FID (Gaz kromatografisi-Alev iyonizasyon dedektörü) cihazında analiz edilmiştir. Pichia kudriavzevii olan A10 ve A20 izolatlarının metil ester (biyodizel) verimi sırasıyla A10 izolatı için %96,17 ve A20 izolatı için %95,85 olarak bulunmuştur. A10 ve A20 kodlu izolatların yağ asidi dağılımı A10 kodlu izolat için palmitik asit (%15,26), palmitoleik asit (%9,99), stearik asit (%9,72), oleik asit (%46,36) ve linoleik asit (%13,25) olarak bulunmuştur. A20 kodlu izolatta ise palmitik asit (%12,48), palmitoleik asit (%11,42), stearik asit (%10,97), oleik asit (%44,39) ve linoleik asit (%13,43) olarak bulunmuştur. Çalışmada kullanılan maya lipitlerinin (Pichia kudriavzevii) çoğunluğunu C16 ve C18 metil esterlerinin oluşturması biyodizel üretiminde hammadde olarak kullanılabilme potansiyelini göstermektedir.
Yeasts are microorganisms that have the potential to be used as a raw material in the production of biodiesel, as they produce high amounts of lipids in the cell under certain conditions. The aim of this study is to determine the biodiesel production potential of yeasts from food-borne microorganisms that have been little studied. In accordance with this purpose; Among the isolated yeasts, it was aimed to discover yeast isolates with the highest lipid yield under nitrogen deficiency stress and suitable fatty acid composition for biodiesel production. Eight yeast isolates obtained from food were incubated at pH 5.5, 30 °C, 100 rpm (shaking speed) and 24 hours in both optimal medium (in the presence of nitrogen) and lipid production medium (lack of nitrogen) and compared in terms of lipid yield and % lipid content. A10 and A20 coded isolates with the highest values were defined as Pichia kudriavzevii species. Nitrogen deprivation stress was applied to A10 and A20 coded isolates, which were identified as species, at 30 oC temperature, 100 rpm and different pH (6-8-10) values for 72 hours. It was observed that both isolates reached the highest lipid content at pH 10 (A10 40%, A20 27%). The A10 coded isolate reached the highest lipid yield at pH 10 with 431.86 mg/day, while the A20 coded isolate reached the highest lipid yield at pH 6 with 219.37 mg/L/day. The methyl esters obtained by transesterification of lipids extracted from yeast cells were analyzed in the GC-FID (Gas chromatography-Flame ionization detector) device. The methyl ester (biodiesel) yield of A10 and A20 isolates of Pichia kudriavzevii was found to be 96.17% for A10 and 95.85% for A20 isolate, respectively. Fatty acid distribution of isolates coded A10 and A20 For isolates coded A10, palmitic acid (15.26%), palmitoleic acid (9.99%), stearic acid (9.72%), oleic acid (46.36%), and linoleic acid (13.25%) was found. Palmitic acid (12.48%), palmitoleic acid (11.42%), stearic acid (10.97%), oleic acid (44.39%) and linoleic acid (13.43%) were found in the A20 coded isolate. The fact that C16 and C18 methyl esters form the majority of yeast lipids (Pichia kudriavzevii) used in the study shows the potential to be used as a raw material in biodiesel production.
