Histamin gibi bazı biyojen aminler çeşitli gıdalarda bulunurlar. Biyojen aminlerin yüksek derişimleri doğrudan veya dolaylı toksik etkiye sebep olabilmektedir. Bu nedenle eser miktardaki histamin tayini önemlidir. Bu çalışmada Kare Dalga Sıyırma Voltametrisi (KDSV) ve Dönüşümlü Voltametri (DV) yöntemleri kullanılarak histaminin elektrokimyasal davranışı çalışılmıştır. Cu(II) iyonları ve histamin varlığında Cu(II)-histamin kompleksine ait yeni bir pik gözlenmiştir. pH 10'da (BR tampon çözeltisinde) Cu(II)-histamin kompleksine ait indirgenme piki, optimum şartlar altında standart histamin eklemelerine karşılık kantitatif artışlar göstermiştir. KDSV yöntemi ile -388 mV'da gözlenen keskin pik kullanılarak eser miktardaki histaminin doğrusal çalışma aralığı 1,0 ?M ? 8,0 ?M ve 3,0 x10-5 mol L-1 ve 9,0 x10-5 mol L-1 olarak tespit edilmiştir. Gözlenebilme sınırı ve tayin sınırı sırasıyla 0,33 ve 1,0 ?M olarak elde edilmiştir. Önerilen metot çeşitli balık numunelerindeki histamin tayini için başarıyla uygulanmıştır. Bazı inorganik ve organik tuzların histamin tayinine etkisi incelenmiştir. pH 10'da 0.00 mV ile -900 mV [Ag/AgCl (3 mol L-1 NaCl)] aralığında asılı cıva elektrot üzerinde Cu(II)-histamin kompleksine ait dönüşümlü voltamogramı alınmıştır. Biogenic amines such as histamine can be found in various foods. Biogenic amines can cause direct or indirect toxicity when their concentration levels are high. Therefore, determination of trace histamine is very important. In this work, the electrochemical behavior of histamine was studied using the Square Wave Stripping Voltammetry (SWSV) and Cyclic Voltammetry (CV). In the presence of Cu(II) and histamine a new voltammetric peak appears which corresponds to a Cu(II)-histamine complexes. The voltammetric reduction corresponding to the Cu(II)-histamine complex peak at pH 10 (BR buffer solution) showed quantitative increments with the additions of standard histamine solution under the optimal conditions. By SWSV a single, sharp peak at about -388 mV is obtained, allowing trace determination of histamine in the range 1,0 ?M ? 8,0 ?M and 3,0 x10-5 mol L-1 and 9,0 x10-5 mol L-1 with a linear calibration. The limit of detection (LOD) and limit of quantification (LOQ) were obtained as 0,33 and 1,0 ?M, respectively. The proposed method was successfully applied to the determination of histamine in various fish samples. The influences of some other commonly found inorganic and organic salts on the determination were also examined. A cyclic voltammogram of Cu(II)-histamine complex was obtained on a hanging mercury drop electrode at pH 10.0 during the potential scan from 0.00 mV to -900mV vs. Ag/AgCl (3 mol L-1 NaCl). From repetitive cyclic voltammograms, one anodic peak at -328 mV and one cathodic peak on the reverse scan at -430 mV were recorded.
