Bu çalışmada, Ponceau 4R ve Sudan Orange G boyalarının katı faz ekstraksiyonu yöntemiyle zenginleştirilmesi ve UV-Vis spektrofotometresi ile tayini gerçekleştirilmiştir. Bu amaçla deniz süngeri ilk kez kullanılmıştır. Deniz süngerinde adsorbe olan Ponceau 4R ve Sudan Orange G 10 mL pH 4,5 (asetat) tamponu ile elüe edildikten sonra spektrofotometrik olarak tayin edildi. Yöntemin optimizasyonu için pH, örnek ve elüent akış hızları, örnek hacmi, sünger miktarı, HCl derişimi, diğer boyaların etkisi ve matriks etkileri gibi değişik analitik parametrelerin etkisi incelendi. Ponceau 4R ve Sudan Orange G boyalarının geri kazanım değerleri % 95 den yüksektir. Gözlenebilme sınırı Ponceau 4R için 0,002 ?g/L, Sudan Orange G için 1,9x10-4 ?g/L olarak bulundu. Yöntemin doğruluğunu test etmek için Ponceau 4R boyası içeren kuşburnu aromalı içecek tozuna ve Sudan Orange G için toz biber örneğine analit ekleme çalışmaları yapıldı. Yöntem çeşitli toz içecekler ve şeker örneklerinde Ponceau 4R tayini için ve çeşitli biber ve biber salçası örneklerindeki Sudan Orange G tayini için uygulandı.
In this study, a solid phase extraction method for preconcentration of Ponceau 4R and Sudan Orange G dyes prior to their UV-Vis spectrophotometric determinations has been investigated. For this purpose, sea sponge was used for first time. Ponceau 4R and Sudan Orange G adsorbed on sea sponge was eluted with 10 mL pH= 4.5 (acetate) buffer before spectrophotometric determination. The effect of various analytical parameters such as; pH, the amount of Ponceau 4R and Sudan Orange G, sample and eluent flow rates, sample volume, the amount of sponge, HCl concentration, effects of some ions and other dyes were examined for the optimization of the method. Ponceau 4R and Sudan Orange G recovery values were higher than 95%. The detection limits for Ponceau 4R and Sudan Orange G were found as 0.002 ?g/L, 1.9x10-4 ?g/L respectively. In order to test the accuracy of the method analyte addition method is applied to a powder drink for Ponceau 4R and to a pepper for Sudan Orange G. The method is applied to determine Ponceau 4R in various powder drinks and candy samples, and Sudan Orange G in various pepper and pepper paste samples.
