Karminik asitin (KA) damlayan cıva elektrot üzerinde indirgenmesinden yararlanılarak difreansiyel puls polarografisi (DPP) ile karmin gıda boyalarının tayini için duyarlı bir metot geliştirilmiştir. DPP ile pH 2 Britton-Robinson (B-R) elektroliti ortamında -489 mVdaki pikinden yararlanılarak yapılan tayinde pik akımı ile derişim arasında doğrusal olarak çalışılabilen aralık 1x10-6 mol/L - 9x10-5 mol/L olarak tespit edilmiş olup gözlenebilme sınırı 1,6x10-7 mol/Ldır. Önerilen elektrokimyasal tayin yöntemi karmin boyası katılan gıdalardan çilekli süte ve meyveli şekere uygulanmıştır. Meyveli şekerdeki KAnın tayini hem DPP hemde spektrofotometrik yöntemle yapılarak iki metot karşılaştırılmıştır. KAnın pH 2 BR elektroliti ortamında asılı cıva damlası üzerinde Ag/AgCl elektroda karşı 0,0 1000 mV aralığında dönüşümlü voltamogramları (DV) alınmıştır. Alının DV?de -500 mVda bir katodik ve tarama hızı ile belirginleşen üç anodik pik gözlenmiştir. Ayrıca KAnın tayinine bazı organik ve inorganik maddelerin girişim etkileri incelenmiştir
As basis for the development of a sensitive analytical method for the determination of carmine food dye, a study of the differential pulse polarographic (DPP) reduction of carminic acid (CA) on a dropping mercury electrode (DME) was performed. For the analytical DPP method running at pH 2.0 Britton-Robinson (B-R) buffer (peak at ?489 mV), the relationship between the peak current and CA concentration was linear in the range of 1x10-6 to 9x10-5 mol/L with a detection limit of 1,6x10-7mol/L. The proposed electrochemical procedure was successfully applied to the determination of carmine food dye in spiked commercially available strawberry flavored milk. The method was extended to the determination of CA in candies and results were in agreement with that obtained by spectrophotometric comparison method. Cyclic voltammogram of CA in 2.0 B-R buffer electrolyte was obtained on HMDE at pH 2.0 during the potential scan from 0.00 mV to 1000 mV vs. Ag/AgCl. From repetitive cyclic voltammograms, one cathodic peak at about -500 mV and three anodic peaks on the reverse scan at about between -340 mV - -460 mV were recorded. The influences of some other commonly found inorganic and organic salts on the determination of CA were also examined.
